PTEN是一个关键的抑癌基因,其蛋白在肿瘤中经常发生缺失。PTEN蛋白的缺失可由遗传学和表观遗传学因素导致,但在肿瘤中PTEN蛋白的缺失比例常常比其基因突变、缺失比例要高很多,因此PTEN蛋白的降解成为肿瘤中研究的一个热点问题。以往研究发现,PTEN蛋白可受到多种翻译后修饰并转移到蛋白酶体进行降解,也有少量研究发现PTEN可经由溶酶体途径降解,但PTEN溶酶体降解的具体分子机制尚不清楚。
